Necrotic pyknosis is a morphologically and biochemically distinct event from apoptotic pyknosis.

چکیده
Classification of apoptosis and necrosis by morphological differences has been widely used for decades. However, this usefulness of this method has been seriously questioned in recent years, mainly due to a lack of functional and biochemical evidence to interpret the morphology changes. To address this matter, we devised genetic manipulations in Drosophila to study pyknosis, a process of nuclear shrinkage and chromatin condensation that occurs in apoptosis and necrosis. By following the progression of necrotic pyknosis, we surprisingly observed a transient state of chromatin detachment from the nuclear envelope, followed by the nuclear envelope completely collapsing onto chromatin. This phenomenon led us to discover that phosphorylation of barrier-to-autointegration factor (BAF) mediates this initial separation of nuclear envelope from chromatin. Functionally, inhibition of BAF phosphorylation suppressed necrosis in both Drosophila and human cells, suggesting that necrotic pyknosis is conserved in the propagation of necrosis. In contrast, during apoptotic pyknosis the chromatin did not detach from the nuclear envelope and inhibition of BAF phosphorylation had no effect on apoptotic pyknosis and apoptosis. Our research provides the first genetic evidence supporting a morphological classification of apoptosis and necrosis through different forms of pyknosis.

Profiling molecular factors associated with pyknosis and developmental arrest induced by an opioid receptor antagonist and dihydroartemisinin in Plasmodium falciparum
Malaria continues to be a devastating disease, largely caused by Plasmodium falciparum infection. We investigated the effects of opioid and cannabinoid receptor antagonists on the growth of intraerythrocytic P. falciparum. The delta opioid receptor antagonist 7-benzylidenenaltrexone (BNTX) and the cannabinoid receptor antagonists rimonaband and SR144528 caused growth arrest of the parasite. Not...
متن کاملUltraviolet and green light cause different types of damage in rat retina.
PURPOSE To assess the influence of wavelength on retinal light damage in rat with funduscopy and histology and to determine a detailed action spectrum. METHODS Adult Long Evans rats were anesthetized, and small patches of retina were exposed to narrow-band irradiations in the range of 320 to 600 nm using a Xenon arc and Maxwellian view conditions. After 3 days, the retina was examined with fu...
متن کاملA Microspectrophotometric Study of Red Cell Nuclei during Pyknosis
1. A microspectrophotometric apparatus was used to measure nuclear absorption in fixed and stained human bone marrow smears. 2. The validity of the measurements was established. 3. The total and polymerized desoxyribose nucleic acid (DNA) of erythroblast nuclei were measured in relative units, and a gradual loss of DNA from the red cell nucleus during maturation was noted. 4. The loss in DNA is...
متن کاملA Study of Gluco-corticosteroid-induced Pyknosis in the Thymus and Lymph Node of the Adrenalectomized Rat
Pyknotic nuclei, observed in the thymus of steroid-treated rats, are dense, homogeneous, intensely basophilic and Feulgen positive. Under the electron microscope, the image is that of a complete segregation of the chromatin from the nuclear sap producing a margin or crescent of condensed chromatin. Approximately 30% of all small thymocytes appeared to undergo this type of degeneration within 3-...
متن کاملSystemic Histopathology of Infant Rats Exposed to Busulfan
Busulfan is an antineoplastic bifunctional alkylating agent. We previously reported the busulfan-induced systemic histopathological changes in fetal rats and the sequence of brain lesions in fetal and infant rats. In the present study, in order to clarify the nature and sequence of busulfan-induced systemic histopathological changes in infant rats, 6-day-old male infant rats were subcutaneously...
